Mario Maurer lahir di Bangkok, Thailand. Ia bersekolah di St. Dominic School, Bangkok, sebelum melanjutkan kuliah di Jurusan Seni Komunikasi Universitas Ramkhamhaeng.

Mario mengawali kariernya sebagai peragawan dengan menjadi foto model, bintang iklan, dan bintang video klip saat ia masih berusia 16 tahun.
